Description

Brigatinib, which used to be called AP26113, is a reversible dual inhibitor of anaplastic lymphoma kinase (ALK) and epidermal growth factor receptor (EGFR). It works better against the mutant forms of EGFR than it does against the wild-type. 1 It is also selective against nine different mutants of the EML4-ALK fusion gene that are resistant to Crizotinib. This gene is a critical player in the transformation of susceptible lung parenchyma. 2 Brigatinib was made by Ariad Pharmaceuticals, a part of the Takeda Pharmaceutical Company Limited. On April 28, 2017, the FDA approved it.

Dosage

  • The recommended dose of brigatinib for adults is 90 mg daily by mouth for the first seven days. If 90 mg is well tolerated, the dose can be increased to 180 mg daily by mouth.
  • The pills should be taken until the disease worsens or any side effects are found.

Changes To The Dose For Side Effects:

  • If a patient has side effects, the treatment may need to be stopped or put on hold for a while.
  • If there are side effects, the dose may need to be changed as follows: • If the patient took 180 mg at first, the dose could be cut to 120 mg daily.
  • If harmful side effects happen at a dose of 90 mg, the amount can be cut to 60 mg daily.
  • Brigatinib therapy should be stopped if side effects happen with a 60 mg dose.

Side Effects

Common:

Nausea, diarrhoea, tiredness or feeling tired, a headache, and a cough.

Gastrointestinal:

Constipation, stomach pain, vomiting, loss of appetite, and high levels of the pancreatic enzymes amylase and lipase are all symptoms of celiac disease.

Cardiovascular:

Heart rate slowing down, activated partial thromboplastin time lasting longer, and high blood pressure.

The brain and spinal cord:

Not getting enough sleep, peripheral neuropathy, and headaches.

Lungs and lungs:

Breathing problems, cough, interstitial lung disease/pneumonitis, and pneumonia are all signs of this.

Musculoskeletal:

Pain in the muscles, back, joints, and extremities

Precautions

After the last dose of brigatinib, women who could have children should use non hormonal birth control for at least four months, and men should use a condom for at least three months. Treatment with brigatinib may make people unable to have children. Patients should tell their doctors if they have unusual muscle pain, tenderness, or weakness.

Overdose

It is recommended that a patient be observed for the onset of adverse responses in the event of an overdose. Adequate care needs to be given to patients who experience adverse reactions.

Storage

Keep the brigatinib tablets in their original, labelled container at a room temperature of 20°C to 25°C. Keep them away from excess heat and moisture. Keep them out of the reach of children and pets.
